Osteoblast cell adhesion on RBM and sand blasted acid etched titanium surfaces: preliminary results.

Abstract

Evaluate sand-blasted-acid attached and resorbable blast media surfaces with cell culture test. Twenty-four grade-4 titanium discs (12-SLA, 12 RBM) at 5mm diameter were used. Human osteosarcoma (SAOS-2) cells were cultured and counted by Trypan blue staining. Aliquots of 8_L of cell suspension containing 2,000 cells were seeded onto the top of samples placed in the wells. Samples were incubated for one, seven and twelve days. The Cell Titer 96TM Nonradioactivity Cell Proliferation (MTS) assay wasused to determine the cell density on the samples and SEM survey was performed after fixing and gold coating. All experiments were performed twice. At 7th and 12th days, cell proliferation on RBM was higher than on SLA but no meaningful statistical difference was evident. Cell morphology presented similar structure with SEM. Our data demonstrated that RBM surface showed at least equivalent biological response of osteoblast-like SAOS-2 cells than SLA titanium during a short-time cell culture period.
